How to decline a work which is given by your manager?

My manager assigned me some automation testing work but I am more interested in manual testing. I tried but not able to do it .Now he is asking to deny this work in formal mail. I have tried: Tried to debug the code for last two weeks. I think it was caused by: Not having experience in coding area
