Along the horizontal toolbar at the top of the window, you can click and change your stroke and fill colors, "Stroke Weight", the shape of your brush and also "Opacity".
You can also double-click the "Paintbrush Tool" icon in your vertical toolbar to bring up the brush settings.
You can adjust the "Fidelity" and "Smoothness" values by dragging the scrollers to the left and right.
