No, you cannot. Transferring the contents of an ISO file will not allow you to boot up from the device to which you copy the files, as the boot sector will not be affected. You will need to follow the article steps to boot up from your USB drive.

Hello Daniel...Can I use an inserted win7 SP1 DVD to make a USB installer stick?

I recently downloaded and burned successfully a DVD image of Win7 Home Premium SP1 64bit on a different computer (different location). However I forgot to take with me my external DVD drive to re-install this Win7 DVD on my Notebook at home. However, I do have a desktop PC at home with a slow WiFi connection. Can I avoid the re-download of the image, by using the inserted DVD and using the MS DVD/USB tool ?

You should be able to pull that off without a problem. Make an ISO of your disc and use that and you'll be right as rain. If that doesn't work for whatever reason, you'll be best served by downloading the file again.
